KPV (lysine-proline-valine) has no documented side effects in people, because no study among the research behind this page gave it to a person. What exists is a short record from animals and cultured cells, built to show whether an induced inflammation eased rather than to catch harm.
The nearest thing to a toxicity reading is cellular. A 2017 mouse colitis paper reported that its KPV-loaded nanoparticles appeared nontoxic and biocompatible with intestinal cells, and a 2006 rabbit study found corneal cell viability went up, not down, at the concentrations it tested. Both describe a dish over hours, not a body over weeks.
The human material is human cells: skin cells exposed to fine dust in a 2025 laboratory study, where KPV restored their viability. The longest stated exposure in any animal is four days of eye drops in rabbits. Nothing in that span can show what months of use would do, and nothing in it tried to.
No approval for any use is recorded among these sources. A 2026 critical review of peptides in sport, which names KPV among the fragments it covers, lists cardiovascular strain, insulin resistance, disordered blood fats and psychiatric instability as emerging risks across the class, and it warns that products in this supply chain are often mislabeled or contaminated.

What side effects are documented for KPV?

None in a person, and the reason is structural rather than reassuring. Every study among the research behind this page used animals, cultured cells or bacteria, and each one was designed to measure whether an injury the researchers created got better. The closest any of them comes to a harm finding is a 2017 mouse colitis paper, which reported that its KPV-loaded nanoparticles appear to be nontoxic and biocompatible with intestinal cells.1 Read the subject of that sentence with care. It is the particle, tested against gut cells, and it says nothing about how a whole animal fared over weeks.
The risk list that does exist belongs to the wider class: a 2026 critical review of peptides used in sport names cardiovascular strain, insulin resistance, dyslipidemia, and psychiatric instability as emerging risks, dyslipidemia meaning disordered blood fats.2 That review names KPV among the fragments it covers, but it does not tie any of those four risks to KPV in particular. So the honest answer is a short list with nothing on it, and on this compound an empty list records how little was looked for, not how little was found.
Is KPV the safest peptide?

That ranking cannot be made from the evidence, for KPV or for any other compound in this class. A safety ranking needs comparable human exposure for each candidate, and KPV has no human exposure on record to enter into the comparison. The 2026 sport review describes the whole category as marketed as more selective and ostensibly safer alternatives to anabolic steroids, and the word ostensibly is the authors' own.2
The appeal of KPV in particular usually rests on where it comes from. A 2025 skin cell study describes it as an endogenous peptide derived from α-melanocyte-stimulating hormone, a hormone the body already makes, and that origin is the usual root of the idea that it must be gentle.3 Being a piece of a natural hormone says nothing about the amount, the route, the purity or the length of exposure, and those are the variables a safety record has to cover. The same sport review concludes that until longitudinal data clarify their safety and prevalence, peptide use in competitive and recreational settings alike should be considered high-risk.2 Safest is a comparison with no second column to read from.
Who should not take KPV peptide?

No group can be named from evidence, because exclusion lists come from human trials and the research behind this page contains none to draw from. What the animal and cell record can do is point to the people for whom the open questions are sharpest, and two stand out.
The first is anyone whose immune system is already turned down, by illness or by medicine. KPV was studied precisely because it calms inflammatory signalling, and the evidence for that is set out in the section on the immune system below. Adding a second brake to a system that is already braked is an obvious question, and none of the studies covered here asked it, in any species.
The second is the young, since the 2026 sport review notes growing uptake among recreational gym-goers, including younger individuals, and adds that prevalence studies are lacking.2 Pregnancy, childhood, kidney disease and old age do not appear anywhere in this record, in any animal model either. Absence from the record is the finding for every one of those groups, and it cannot be read as clearance.
Can you take KPV every day?

Daily use has been examined only in the narrow sense that a few animal experiments administered KPV repeatedly over a short, defined period. The clearest schedule is a 2006 rabbit study, in which animals were topically treated with KPV 1, 5 or 10 mg/ml, two drops four times in a day, for 4 days.4 Those were eye drops applied to a deliberately abraded cornea, and observation ended once the surface had regenerated. A 2022 diabetic mouse wound study took the other approach, building a layered dressing in which KPV was firstly released within 3 day before a growth factor followed it.5
Among the studies behind this page that state a duration, the longest is the four days of eye drops in rabbits. Continuous daily exposure over months in a person is a different situation altogether, and repeated use across weeks is precisely where any cumulative effect would become visible. That has not been measured, so daily use sits wholly outside what was tested rather than at the far edge of it. The amounts and schedules the studies did use are laid out on the KPV dosage page.
Does KPV dampen the immune system?

In cells and in mice it dampens inflammatory signalling, which is the effect every study was looking for. In a 2003 cell study, every KPV analogue tested dose dependently inhibited nitric oxide output in mouse immune cells, and nitric oxide is one of the signals those cells send out when they are inflamed.6 A 2025 review puts KPV in a group of peptides that work by downregulating the nuclear factor kappa B (NF-κB) pathway, modulating cytokine release, and restoring homeostasis, where NF-κB is the main on-switch for genes that drive swelling.7
The most direct line comes from a 2024 bowel disease paper, in which the nanocarriers induced (or did not induce) immunosuppression depending on the presence (or absence) of a hyaluronan-KPV functionalization.8 In that system the KPV coating was the switch that turned immune suppression on. A 2024 mouse colitis study ran KPV in its own group next to FK506, a standard medicine that holds down the immune system, and its combined particle restored the seals of the gut lining to levels surpassing those observed in the KPV and FK506 groups.9 Damping inflammation is the intended effect, and it is also why infection risk is the obvious unmeasured question. What that braking does to an animal fighting a real infection has not been asked anywhere in this record.
Can you get KPV in pill form, and does the form change the risk?

Swallowed forms exist in the research, but none of them is a plain capsule of KPV, and that changes what any safety reading covers. The oral version that performed best in mice was a conjugate integrating a hydrophilic polyethylene glycol segment, a reactive oxygen species (ROS)-responsive hydrophobic self-immolative module, and a hydrolyzable scaffold, which is to say a carrier designed to fall apart only where tissue is inflamed.10 In mice with colitis it came with enhanced efficacy even at a 20-fold lower dose than free KPV, which shows how much of the outcome belonged to the carrier.10
A 2017 oral system went through two layers, with the nanoparticles encapsulated in a hydrogel (chitosan/alginate) before the mice swallowed them.1 Every one of those materials is part of what the animal received, and the results describe the whole package. A capsule of plain KPV powder is a different exposure from any of them, and none of the studies covered here examined it for harm. What did the cell tests show about toxicity?

The direct readings of KPV against living cells point away from damage, at the concentrations tested. In a 2006 study, rabbit corneal epithelial cells in culture were exposed for 1, 6 and 24 h to different KPV concentrations (0.1, 1 and 10 microM).4 Cell viability was stimulated by 1 and 10 microM concentrations, so the count of living cells rose rather than fell.4 In human skin cells damaged by fine airborne dust, treatment with 50 μg/mL of KPV restored cell viability and reduced IL-1β secretion, IL-1β being one of the main inflammatory messengers.3
Those results share a limit that matters more than their direction. A culture dish contains a single cell type for hours or a day, with no liver, no kidney and no circulating immune system to register a problem elsewhere. A clean reading in a flask is measured against that single cell type, and it is a long way from a clean reading in an animal, let alone in a person.
Does KPV affect blood sugar, blood fats or the liver?

One experiment touched these organs, and the readings moved in the direction its authors were hoping for. In a 2026 high-fat diet mouse study, oral administration of KPV alleviated body weight gain, white adipose tissue expansion, liver mass increase, and obesity-associated dyslipidemia, including elevated plasma total cholesterol levels.11 Those are markers of a diet-induced change in fattened mice rather than a toxicity panel, so they show the peptide pushing against an injury, not what it does to a healthy liver.
Blood sugar is the larger gap. Insulin resistance appears on the 2026 sport review's list of emerging risks for the class, and no study behind this page measured glucose or insulin in an animal given KPV. Kidney function is absent from the record in the same way. So the honest position on organ effects is one mouse experiment on liver mass and blood fats, read as a benefit by its authors, and nothing at all on blood sugar or the kidneys.
Will KPV darken skin the way its parent hormone does?

The research says it should not, and the reason is specific. A 2019 review of melanocortin peptides in wound healing reports that short fragments such as KPV have been found to possess anti-inflammatory effects but to lack the pigment-inducing activity of α-MSH.12 A 2003 cell study points to why: the fragment and its analogues did neither bind to MCRs nor stimulate cAMP, MCRs being the melanocortin receptors through which the parent hormone works.6
There is a complication in the newest work, because a 2026 vitiligo study used KPV as a homing tag, building lysine-proline-valine (KPV)-modified deformable liposomes to reach melanocytes, the skin's pigment cells, in a mouse model.13 Steering particles toward pigment cells and changing skin colour are different things, and the vitiligo work measured the first. Skin colour in an animal given KPV was not measured in any study behind this page, so the answer rests on mechanism rather than on observation.
What does KPV interact with?

One real interaction is documented, in rabbits, and it runs through nitric oxide. In the 2006 eye study, pre-treatment with L-NAME, a compound that blocks nitric oxide production, inhibited the facilitating effect of KPV on corneal epithelial wound healing in rabbit eyes.4 Block that pathway and the effect on the wound went with it.
Every other pairing on record was designed by the researchers rather than observed. A 2024 bowel paper built nanoparticles loaded with teduglutide for combination therapy in IBD, teduglutide being a separate gut-acting medicine and IBD meaning inflammatory bowel disease.8 A 2022 diabetic mouse wound film carried an anti-inflammatory tripeptide (KPV) and epidermal growth factor (EGF) in separate layers.5 Each part had a defined job, and the experiments tested the package. That says nothing about how KPV behaves beside the medicines a person already takes, and none of the studies covered here combined it with BPC-157, GHK-Cu or TB-500. Is KPV banned in sport?

The research behind this page does not settle the listing question. The 2026 sport review names KPV only as an example, listing synthetic fragments (e.g., Frag 176-191, KPV) among the peptides it covers.2 It describes testing across the whole class, noting that detection is hard because these peptides look like the body's own hormones and clear quickly, and it does not say where KPV sits on any prohibited list. An athlete needs the current list itself rather than a review of the market, because a review that names a compound as marketed is not evidence that the compound is either permitted or banned.
Does the vial carry risks that KPV itself does not?

Yes, and these are the risks the research is least able to speak to. The 2026 sport review warns that the largely unregulated supply chain exacerbates these dangers, as products are often mislabeled or contaminated.2 A 2026 analysis for compounding pharmacists notes that compounded peptides have quickly moved from a niche topic to a growing area of interest among patients and prescribers, and it sets out the concerns raised during a United States advisory review.14
Stability is the quieter problem. A 2021 rat colitis study states that KPV solution is very unstable when rectally administered, which is why its authors built a gel to hold it.15 A 2022 rat paper reported that its gel kept KPV retaining its bioactivity for a longer time under high temperature conditions, which implies the plain peptide did not hold up as well.16 A mislabelled vial, a contaminated one and a solution that has broken down are all problems of the material in hand, and an animal study cannot vouch for any of them.
How long does the KPV safety record run?

Hours to days, in every animal that states its timing. The oldest experiment, from 1991, measured mouse ear swelling 3 and 6 h after application of the irritant.17 A 2021 rat study of mouth ulcers caused by chemotherapy recorded that its KPV gel adhered to the wound surface for 7 hours when administrated to the gingival mucosa of rats, the gingival mucosa being the gum lining.18 The rabbit eye study ran four days, which is the long end of the record.
The 2026 sport review describes peptides of this kind as experimental substances with poorly defined long-term risks.2 For KPV that description is generous, since long-term risk is not poorly defined so much as unmeasured. A safety record covers the exposure it watched, and on this compound that is less than a week in any animal that reports its timing, rather than the months a person might use it.
Why is the KPV harm record this thin?

Because most of the research is about building delivery systems, and KPV is often the cargo rather than the subject. A 2022 rat colitis paper describes KPV (Lys-Pro-Val) as a model drug that its new hydrogel could capture.16 The 2021 mouth ulcer paper put it in identical terms, using tripeptide KPV as a model drug inside its gel.18
A 2023 review of the melanocortin system in bowel disease notes that numerous experiences have been accumulated in mouse models of colitis, which is where most of the work sits.19 When a molecule is the cargo, the experiment is built to show that the vehicle works and that the disease model improves. Harm to the animal is a different question, and this literature has mostly not been asked it. That is why the record is short, and why short should be read as untested rather than as cleared.
What we don’t know
The gaps in the evidence matter as much as the findings.
