Epitalon (also spelled epithalon) is a research compound studied for ageing, and almost all of what is known about it comes from rodents, fruit flies and cells in a dish. Most of that work was done by one research group in St Petersburg.

The best-known findings are in two separate places. In human cells grown in a dish, it switched on telomerase, the enzyme that rebuilds the protective caps on chromosomes. In whole animals, the result was narrower: in one mouse study it did not change average lifespan but extended maximum lifespan by about 12%.

Very little of this has reached people in the form sold as Epitalon. The long human studies, including one that followed 266 older people for six to eight years, used epithalamin, the pineal-gland extract that Epitalon was designed to copy. A cell result, an animal lifespan result and an extract trial in people are three different kinds of evidence, and none of them stands in for the others.

No approval for Epitalon in any country is documented among the research behind this page. A 2025 review notes that it is still uncertain whether the effects described so far are the only ways the compound works.

What is Epitalon?

review

Epitalon is a short synthetic peptide, a chain of four amino acids, made in a laboratory and studied mainly as an anti-ageing research compound. A 2025 review describes it as a tetrapeptide, Ala-Glu-Asp-Gly (AEDG), which was synthesized based on the amino acids composition of Epithalamin, a bovine pineal gland extract.1 The pineal gland is the small gland in the brain that makes melatonin, the hormone that tracks night and day.

The order of events matters. The peptide was built first, from a recipe read off the extract, and only later looked for in the gland itself. A 2017 paper from the developers reports that peptide AEDG was detected among the tetrapeptides of the epiphysis polypeptide complex, using mass spectrometry.2 So it is a copy of one part of an animal extract, and it was named for that extract rather than discovered as a hormone in its own right.

Are epithalon and Epitalon the same?

animal model

Yes. Epitalon, epithalon and epithalone are spellings of one molecule, and the papers use them interchangeably. AEDG, the four-letter code for its amino acids, is a fourth name for the same thing.

The name that is not interchangeable is epithalamin. A 2007 rat study draws the line clearly, describing epithalamin (polipeptide preparation from bovine brain) and its active fragment, epitalon (Ala-Glu-Asp-Gly).3 One is a mixture of many peptides taken from cattle tissue, and the other is a single small peptide built in a laboratory.

A 2022 mouse egg-cell study from outside that group puts it the same way, describing a synthetic short peptide made of four amino acids, based on a natural peptide called epithalamion extracted from the pineal gland.4 Results for the extract cannot simply be relabelled as results for Epitalon, and this page names which one each study tested.

What does Epitalon do to the body?

animal model

None of the studies we could find followed what it does to a whole, healthy person over time, so the honest answer is a list of what it did in particular models. The developers' 2002 review reports that Epitalon increases the lifespan of mice and fruit flies and restores the circadian rhythms of melatonin and cortisol production in old rhesus monkeys.5 Cortisol is the main stress hormone, and in a healthy body both it and melatonin rise and fall on a daily cycle.

A second recurring claim is that it acts as an antioxidant, meaning it limits the chemical damage caused by reactive oxygen. In old rats, the same group reported that the pineal peptides enhance the antioxidant defense system, measured in blood serum, liver and brain.3

Read those together and a pattern shows. The effects were seen in old animals, with control groups, inside one research programme. That is real evidence about those animals, and it is not the same thing as evidence about what the compound does in a person who buys it. The outcome-by-outcome detail is on the Epitalon benefits page.

Has Epitalon been tested in people?

in vitro

Only a little, and the published abstracts give few details. Two studies in the material behind this page gave the synthetic peptide itself to people.

The first is in eye disease: the 2002 review says Epitalon improves the visual functions in patients with pigmental retinal degeneration, an inherited condition that slowly destroys the retina.5 The second measured melatonin. A 2020 paper reports that AEDG peptide increases in 1,7 times the 6-sulfatoxymelatonin (6-SOMT) excretion in the urine of middle-aged people, a breakdown product used to track how much melatonin the body makes.6

Neither abstract states how many people took part, how they were chosen, or whether a comparison group was given a dummy. That does not make the findings false. It does mean they cannot be weighed, and a reader should treat them as early reports rather than trial results. Several other papers used human cells instead. One studied the peptide in cultured lymphocytes of persons aged 76-80 years, white blood cells taken from older volunteers and kept in a dish, which is cell work rather than a test in a living person.7

What did the epithalamin trials in older people report?

human RCT

The large human numbers that circulate online come from the extract, not from Epitalon. The best known is a study that assessed thymic and pineal peptide preparations in 266 elderly and older persons during 6-8 years, with the preparations given for the first two to three years.8 It reported a death rate lower by 1.6-1.8-fold in the Epithalamin-treated group, compared with controls.8

A smaller randomised study gave 39 coronary patients regular courses of epithalamin on top of their usual care, while 40 coronary patients received basic therapy alone.9 It reported a significantly lower mortality in the group of patients treated with epithalamin in parallel with basic therapy.9

These are the strongest human results in the whole literature on this family of compounds, and they come with two limits a reader cannot see from the headline. Both were run by the research group that developed the preparation, and both tested the cattle extract. Whether a synthetic four-amino-acid copy does the same thing in people is a different question, and it has not been answered in a trial of that size among the studies we could find.

Does Epitalon really lengthen telomeres?

in vitro

In cells grown in a dish, yes, and that result has now been reported by more than one laboratory. Telomeres are the protective caps on the ends of chromosomes, and they shorten each time a cell divides. Telomerase is the enzyme that can rebuild them.

The original 2003 experiment added the peptide to human fetal fibroblasts, a connective-tissue cell that normally lacks telomerase, and recorded expression of the catalytical subunit, enzymatic activity of telomerase, and telomere elongation.10 A 2025 study from an independent team reported dose-dependent telomere length extension in normal cells through hTERT and telomerase upregulation, where hTERT is the working part of the enzyme.11

That is a cell result, and it should be read as one. Longer telomeres in a dish say nothing on their own about longer life in an animal or a person, and the lifespan experiments are a separate body of work. The independent study also recorded what happened in cancer cells, which belongs on the Epitalon safety page rather than here.

Does Epitalon make animals live longer?

animal model

In some experiments, and the details are less dramatic than the popular claim. The clearest test is a mouse experiment that continued until every animal had died. Female mice injected for life showed that treatment with Epitalon did not influence food consumption, body weight or mean life span of mice.12 What changed in these mice was at the far end of the curve: it increased by 13.3% the life span of the last 10% of the survivors and by 12.3% the maximum life span, compared with controls.12

That distinction is the whole story. Average lifespan describes the typical animal, and it did not move. Maximum lifespan describes the final few survivors, which makes it a considerably noisier statistic. There were 54 mice in each group, so the top tenth of survivors amounted to roughly five animals per arm.12 The experiments in rats, other mouse strains and fruit flies vary in similar ways, and they are laid out individually on the Epitalon benefits page. None of these lifespan experiments measured telomeres in the animals whose survival was recorded, so the two findings sit side by side rather than one explaining the other.

Who did the research?

human RCT

Most of it comes from one programme. A 2003 monkey paper notes that epithalamin and epitalon were both developed in the St. Petersburg Institute of Bioregulation and Gerontology (Russia).13 Vladimir Khavinson, who led much of that work, is an author of at least 15 of the 25 papers cited on this page. Partner institutions joined some of the human work, including the Institute of Gerontology of the Ukrainian Academy of Medical Sciences (Kiev).8

That is worth knowing, and it is not an accusation: a group that develops a compound is usually the group that studies it first, and many of these experiments used control groups and reported numerical results. The limitation is replication. When the same institution designs, manufactures and evaluates a compound, independent laboratories provide the check that is still largely missing.

A handful of studies from other teams have now appeared, mostly in cells. They include the 2025 telomere work above, a 2025 study in human retinal cells, and two studies of mouse and cattle egg cells grown in a dish. They are small, and none of them is a lifespan or human study.

What is Vilon peptide, and how is it related?

animal model

Vilon is a sibling compound from the same programme, and it often appears in the same experiments. A 2002 mouse study names it as Vilon peptide (Lys-Glu), a two-amino-acid peptide, and tested it alongside Epitalon.14 That abstract prints Epitalon's last amino acid as Glu rather than Gly, an error worth noticing in a literature where the sequence is the whole identity of the compound.

The developers describe a family of short synthetic peptides, which they call cytogens, including Cortagen, Epithalon, Livagen, and Vilon, each said to act on its own tissue.15 Vilon was matched to the thymus, an immune gland, and Epitalon to the pineal gland.

The two do not behave alike. In mice bred to develop breast tumours, treatment with Vilon produced significant negative effects when compared to the control group, including more breast cancer, while Epitalon reduced tumour numbers in the same experiment.16 Sharing a developer is not the same as sharing a record.

Can Epitalon be taken with other peptides?

human RCT

Combinations have been tested only rarely, and almost never with the synthetic peptide in people. In mice, combined treatment with Vilon and Epithalon changed expression of 144 clones, more genes than either peptide alone.17 That shows the pair did something different together, measured as changes in which genes were switched on in heart tissue. It does not show whether the change helped or harmed.

The only human combination data concern the extracts. The 266-person study above gave thymalin, a thymus extract, together with epithalamin to one group, and reported a death rate lower by 2.5-fold in the patients treated with Thymalin plus Epithalamin, compared with controls.8 That is a result for two cattle extracts given by the same clinic, which is not the same as a result for Epitalon combined with anything sold today.

No study among the research behind this page tested Epitalon together with the compounds it is most often paired with online, such as other longevity peptides. What is documented about mixing it with medicines is set out on the Epitalon safety page.

Where does Epitalon sit among peptides studied for ageing?

review

No research compound can honestly be called the top choice for ageing, because none of the studies we could find tested one against another for that purpose. What can be compared is the kind of evidence each one has. A 2026 review of peptides in gerontology sorted nine compounds by what they target, and placed Epitalon under telomere biology (epitalon).18

That label is accurate, and it also shows how Epitalon differs from its neighbours, because the reasoning behind it rests on cell experiments and animal survival curves. Several other ageing peptides have a different evidential foundation. SS-31 has been through formal drug trials in rare inherited disease, as set out on the SS-31 overview, while MOTS-c is a peptide the body makes for itself and has mostly been measured rather than given, as explained on the MOTS-c overview. NAD+ is not a peptide at all, and the page on the NAD+ overview covers a separate literature on cell energy.

Read the list as a map of where each compound's evidence sits, rather than a ranking. The comparison the published work actually supports is between the species, the outcome measured and the size of each study.

How is Epitalon given in studies?

animal model

Most animal studies injected it under the skin, and the monkey studies injected it into muscle, but researchers have also tried less common routes. One rat study chose intranasal infusions, describing them as a noninvasive way to deliver this peptide to CNS hypassing the blood-brain barrier.19 The central nervous system is the brain and spinal cord, and the blood-brain barrier normally keeps many molecules out of it.

Another gave it by mouth, reporting that peroral administration of Epithalon to male and female Wistar rats changed the activity of digestive enzymes along the gut.20 That finding is about the gut itself, and it does not show that a swallowed dose reaches the rest of the body.

The amounts, schedules and course lengths behind each route are on the Epitalon dosage page. They are study parameters for particular animals, and none of them was designed as an amount for a person.

What is documented about harm?

animal model

Very little, because almost none of the studies behind this page was designed to look for it. The most direct statement comes from a long mouse experiment. In older mice given small injections five times a week for six and a half months, long-term exposure to Epitalon in small doses did not show any toxic effect.21

That is not a human safety record. It is a finding in one strain of mouse at a tiny amount. Even a 2025 cell study that reported a benefit added that more mechanistic investigations are needed to confirm Epitalon's benefits and safety.22

A few animal and cell findings point in less comfortable directions, and the cancer-cell telomere result belongs among them. Each of those, along with what is known about pregnancy, hormones and mixing with medicines, is gathered on the Epitalon safety page.

animal model

No approval for Epitalon in any country is documented among the research behind this page. Its parent extract is a different case. A 2005 monkey study calls Epitalon a synthetic analogue of the pharmacopoeia drug epithalamin, where a pharmacopoeia is an official national list of medicines, though the abstract does not say which country's list.23

That difference is easy to lose. An extract listed as a medicine says nothing about the legal status of a synthetic copy of one of its parts, which was made later and carries its own name. Nor does it tell a reader anything about what is in a vial bought under either name.

A 2015 drug-testing report covers the identification of the small research tetra peptide Epitalon in two illegal pharmaceutical preparations, and that finding is examined on the Epitalon safety page.24 The status of Epitalon in sport is not addressed by any source we could find.

How is Epitalon thought to work?

review

The short answer is that nobody is sure, and the reviews say so. The 2025 review lists effects on melatonin synthesis, immune signalling and several enzymes including telomerase, then states that it remains uncertain whether these are the sole mechanisms of action of this compound.1

The developers' own theory is that very short peptides act directly on genes. In a 2003 paper they reported that sequences of nucleotide pairs that can serve as binding sites for tetrapeptide Epithalon were identified in the promoter regions of genes including telomerase, the stretches of DNA that switch a gene on.25 That was a hypothesis built from analysing sequences, not a measurement of the peptide binding DNA inside a living cell.

The same 2025 review adds that the quantity of physico-chemical and structural investigations of this peptide remains quite limited, meaning basic chemistry such as its shape and stability has had little study.1 The deeper gene and chromatin work is on the Epitalon benefits page.

Regulatory status

No approval for Epitalon in any country is documented among the research behind this page · epithalamin, the pineal extract it was modelled on, is described in one paper as a pharmacopoeia drug · the long human follow-ups used epithalamin, not Epitalon

What we don’t know

The gaps in the evidence matter as much as the findings.

  1. 01Whether Epitalon itself does anything measurable in people under controlled conditions. The two human reports among the research behind this page do not state their size or whether a comparison group was used.
  2. 02Whether the epithalamin results carry over. The six-to-eight-year mortality findings tested a cattle pineal extract, and no study we could find has repeated that design with the synthetic peptide.
  3. 03Whether longer telomeres in cells translate into anything in a whole body. The cell results and the animal lifespan results come from separate experiments, and none of the lifespan studies measured telomeres.
  4. 04How well the findings hold up outside one research programme. Independent work so far is in cells and egg cells, not in animal lifespan or in people.
  5. 05What happens over years of use in a person, at any amount. The longest controlled exposures among these sources are in mice.
  6. 06What is in a vial sold under the name. A 2015 report identified Epitalon in illegal pharmaceutical preparations, and nothing in this literature characterises material sold outside research.
