Argireline: what the research measured

Argireline (acetyl hexapeptide-8) has been measured to soften fine wrinkles around the eyes and to cut water loss from the skin, in small trials lasting four weeks to two months. When the same serum was tested with and without it on the two halves of each face, the difference was not significant.

The strongest result is a 2013 placebo-controlled trial in 60 Chinese volunteers, where the clinicians' overall wrinkle grading gave the peptide group a 48.9% efficacy rate against 0% for placebo, and silicone casts of the skin showed less roughness. A 2017 trial in 24 volunteers recorded smoother skin and less water loss.

The collagen benefits often quoted come from aged mice, zebrafish and skin cells, not from human faces. In 24 people with eyelid spasm, adding the peptide to Botox injections stretched the time before symptoms returned from 3.0 to 3.7 months on average, a trend rather than a significant result.

Many positive results come from serums that combine the peptide with four or five other actives, so they cannot say what the peptide did by itself. It is sold as a cosmetic ingredient, not as a treatment for any condition.

What does Argireline do for your face?

In people, two things have been measured: how wrinkles around the eyes look and feel, and how much water the skin loses. The 2013 placebo-controlled trial graded the appearance of peri-orbital lines using Daniell's classification and Seeman's standard1, two visual grading scales. A 2017 randomised trial measured skin microtopography and transepidermal water loss2, microtopography being the fine surface texture of the skin and water loss being a common measure of how well the skin barrier holds moisture. Everything else on this page, from collagen to scars, was measured in animals, cells, or small uncontrolled groups. A 2025 review summed up the human and animal results as suggesting it may reduce wrinkle depth, improve skin elasticity, and enhance hydration3, and the word may is doing real work in that sentence. The sections below take each outcome in turn and say what kind of evidence stands behind it.

Can Argireline undo wrinkles you already have?

In one placebo-controlled trial, existing lines around the eyes looked and measured somewhat better after a month. In the 2013 trial in Chinese volunteers, the total anti-wrinkle efficacy in the argireline group was 48.9%, compared with 0% in the placebo group1, a figure from the clinicians' visual grading. On silicone casts of the treated skin, the parameters of roughness were all decreased in the argireline group of trial volunteers1, which is the harder measurement because a machine read the casts. A 2018 study cites efficacies up to 48% upon 4 weeks of twice daily treatment4, which appears to be the same result repeated rather than a second trial. The same Chinese group also claims it can reduce the degree of existing facial wrinkles and work against their development5, but that paper's own experiment was in mice. So the human answer rests on one month-long trial with about 45 people on the active cream. Softer-looking lines after four weeks of use is not the same as wrinkles undone, and no trial among these sources checked whether the change outlasted the cream.

What happened when the peptide was tested against its own serum?

It made no measurable difference, which is arguably the most important individual result on this page. In the 2023 study, Argireline was included in the serum applied to the right side of the face6, and the left side got the identical serum without it. Neither the 19 participants nor the researchers knew which side was which until the analysis was finished. Both sides improved a little in the participants, but comparing them, neither demonstrated a significant reduction in wrinkle score (p=0.829)6, and a p-value that high means the two sides were, for practical purposes, the same. This design removes the main problem with the positive trials, which compared the peptide cream against a placebo cream with a different make-up, or against nothing. It is small, and it used a camera system rather than silicone casts, so it does not cancel the 2013 result. It does mean the case for the peptide doing something on its own, beyond the cream that carries it, has been tested once in a clean way and did not hold up.

How long does it take for Argireline to work?

The trials did not measure onset; they measured at fixed visits, so the earliest visit is the best available answer. In the 2017 trial, the difference between the hexapeptide group and the group using neither peptide was already significant after 20 days2, on one measure of surface texture. The 2013 multicentre study assessed its volunteers after 4 weeks of twice-daily use7. In the 2019 microneedle patch trial, improvements were recorded on day 29 after application8. A five-ingredient serum tested in 2026 improved static wrinkle appearance within the first week in its trial participants9, but that serum's speed cannot be credited to the peptide alone. Read across, the visits cluster at three to four weeks, which is why that is the figure most often quoted. It describes when someone looked, not when anything started to happen.

Does Argireline help skin hold moisture?

There is a measured signal in one human trial, and supporting results in animals and lab skin. The 2017 trial reported a significant decrease in TEWL with acetyl hexapeptide-3 treatment2, TEWL being transepidermal water loss, the rate water escapes through the skin. Less water loss usually means a better-sealed outer layer. In zebrafish, a small fish used as a laboratory animal, the peptide reduced water loss from the tail fin after a salt challenge10. In ex vivo human skin, a peptide-loaded film enhanced skin hydration and reduced scaling after 48 h11. The microneedle trial found hydration improved in all groups, including the patch with no added ingredient8, which shows how easily a carrier produces the same result. Moisture is a reasonable thing for a peptide cream to help with, and it is also what almost any cream helps with, so it says little about the peptide's designed purpose.

Does Argireline build collagen?

Partially, in mice and in isolated skin cells, and the human evidence is missing. In aged mice treated twice daily for 6 weeks, a 2013 study found the amount of type I collagen fibers increased5, type I being the main structural collagen of skin. The companion mouse experiment found type I collagen fibers rose while type III collagen fibers decreased7, so the picture was a shift between types rather than more of everything. In human dermal fibroblasts exposed to UV light, a 2026 study found the peptide was one of four that significantly increased type III collagen levels12. In aged human skin samples in the lab, a 2025 study found applying it significantly increased collagen levels11. The mouse and cell results disagree on type III, and none measured collagen in a living person's face. Each also depends on the peptide reaching collagen-making cells, which is the step a cream struggles with.

Did Argireline help eyelid spasm?

It showed a hopeful trend that did not reach significance. The 2013 pilot's primary outcome was the time to return to baseline on a blepharospasm rating scale after a Botox injection started at the same time as the peptide13, blepharospasm being the condition where the eyelids clamp shut. The patients using the peptide trended toward a longer time to return, 3.7 months against 3.0 months on placebo13. One-third of the active group, 4 of 12 patients, had a considerable extension of symptom control, ranging from 3.3 to 7.1 months13. A trend in 24 patients is a reason to run a bigger trial, which is what happened next. A registered Phase 2 trial of the peptide for blepharospasm is listed as terminated14, with 8 people enrolled, so the larger answer was never delivered among the records covered here. The result supports the peptide as a possible add-on to Botox for this condition, not as a substitute for it.

Does Argireline help scars or oily skin?

The evidence for both is weak, and it is weaker than it looks in the abstracts. A 2020 report on 26 patients used a 10% gel-cream to mask scars, skin tags and other marks15, and it reports that the cream significantly improved the skin values and the self-image expectation of each patient15. The same paper calls itself spontaneous, anecdotal and retrospective, meaning the records were gathered from routine care rather than planned as a test, and it had no comparison group. It measured hydration, elasticity and sebum before and after treatment15, sebum being the skin's own oil. A registered study of a topical form on the cosmetic appearance of oily skin is listed as completed16, with 14 people enrolled, but its record as cited here gives no result. A before-and-after improvement with no control is what a placebo cream can also produce.

Do serums with several actives show what Argireline does?

No, and many of the most positive results come from exactly these products. A 2026 serum combining the peptide with four other actives reported mean clinical scoring from 35% to 69% for various types of wrinkles9 in its trial participants. A 2024 report from practising dermatologists describes a serum containing 2% of the peptide alongside a dipeptide, polyhydroxy acids, niacinamide and seaweed extract17. A 2026 delivery system that also contained a conotoxin peptide improved skin elasticity and wrinkle parameters in exploratory human assessments18. Each of these may well work. But when five ingredients go on together and there is no arm without the peptide, the result describes the formula, and the peptide's share of it is unknown. That is the gap the 2023 split-face study was designed to close, and in that test the peptide's share came out at nothing measurable.

Does better delivery give a better result?

In the few tests that compared the same peptide with and without a delivery aid, the boosted version did better. In a 2025 study, participants using the peptide with a malic-acid ionic liquid saw a greater reduction in wrinkle number, length and area after 28 days than those using the peptide alone19. In a 2026 mouse study, peptide-loaded dissolving microneedles also improved skin elasticity and hydration in a photoaging model20. Among four modified versions of the peptide tested on human nerve cells in vitro in 2018, the one called Arg3 was the most effective at blocking signal release4, Arg0 being the original. These results point in the same direction as the penetration problem described on the Argireline overview: the plain peptide in a plain cream is the weakest version of the idea. They also mean a positive result for a boosted product says little about an ordinary jar.

What has only been shown in animals or cells?

Most of the claimed extras, from antioxidant effects to slower ageing of cells. In zebrafish embryos, the peptide oil restored telomerase activity after damage10, telomerase being an enzyme tied to cell ageing. The same animal study found it suppressed neutrophil counts and inflammatory cytokine levels10, both markers of inflammation. In a 2025 lab and zebrafish study, the peptide combined with peony extract and a penetration enhancer showed strong scavenging of reactive oxygen species, along with collagen production21, though the mixture makes the peptide's own share unclear. In a 2025 zebrafish experiment, the peptide served as the positive control and was outperformed on collagen and elastin genes by a new ellagic acid carrier22. A fish embryo bathed in the peptide is not skin with a barrier, so these findings sit a long way from a face.

Which hoped-for Argireline benefits are still unmeasured?

The one it is sold on, above all. No trial among these sources measured facial muscle activity in people using the cream, so the claim that it relaxes the muscles behind expression lines rests on cell work. A 2025 review states that the peptide's ability to inhibit muscle contraction when applied topically remains incompletely understood3. Prevention is in the same position: the claim that it works against new wrinkles comes from the aged-mouse work, not from people followed over years. Lifting and firmness are claimed for related procedures, such as threads soaked in the peptide, but the 2023 thread paper measured release in the lab and called for a clinical trial. Results for men, for older adults or for darker skin types do not appear in the trials described here. Each of these is a reasonable question, and the answer on each is that it has not been asked of the peptide in a controlled human study among the research behind this page, which is not evidence that it fails.

What we don’t know

The gaps in the evidence matter as much as the findings.

  1. 01Whether the peptide adds anything beyond its base. The only study that held the base constant on the same faces found no significant difference.
  2. 02Whether any benefit lasts once use stops. The trials behind this page measured during use, at four weeks to two months, and did not follow people afterwards.
  3. 03Whether the collagen changes seen in aged mice and in skin cells happen in human skin from a cream.
  4. 04Whether it does anything for facial muscle movement in a person. None of the trials covered here measured how the muscles themselves moved.
  5. 05What it does for men, for older adults or for different skin types. The trials enrolled small, mostly female, mostly East Asian groups.
  6. 06Whether the eyelid-spasm trend is real. It came from 24 patients, it was not statistically significant, and a later registered trial was terminated.
