Kisspeptin comparisons and stacks

Kisspeptin has been compared head to head in people exactly once, against a longer-acting version of itself, and never against PT-141 or against hCG in men in any study among the research behind this page. Most comparisons people search for are between compounds that act at different points in the reproductive chain.

The human comparison, published in 2020, gave 21 women either native kisspeptin-54 or the agonist MVT-602. Luteinising hormone rose to a similar height after both, but the agonist kept brain cells firing roughly twice as long in laboratory slices. In rats, a GnRH analogue raised testosterone without changing sexual motivation, while kisspeptin-10 given up the nose did the reverse.

Against hCG, the claim is specific and comes from fertility medicine. A 2022 review states that kisspeptin can induce egg maturation in IVF in a way that can reduce the risk of ovarian hyperstimulation seen with hCG. That is a statement about women having egg collection, not about men or testosterone.

No kisspeptin product is recorded as approved among these sources. The one approval on this circuit is for a drug that blocks a neighbouring receptor, which is a different molecule doing the opposite job.

What has kisspeptin actually been compared against?

Four things, and only one comparison involved people. The human one set native kisspeptin-54 against the agonist MVT-602 in women, and it has its own section below. In rats, the comparison groups received saline, buserelin acetate, a GnRH analogue, at 20 μg, and kisspeptin-10 by two routes.1 The same study also included a group given Yoquimbine 200 µg, the spelling the abstract uses for yohimbine.1 The abstract reports no result for that group, so that comparison was set up in the design and then left out of the summary a reader can see. The Leydig-cell study compared a range of kisspeptin-10 doses with control rats injected with physiological saline.2 That is a comparison with nothing, which is the most basic kind. Nothing among the research behind this page sets kisspeptin against PT-141, testosterone or hCG in the same experiment.

What to stack with kisspeptin?

No stack appears in any study among the research behind this page, and the compounds that do appear next to kisspeptin mostly act on it rather than beside it. In overweight PCOS rats, a herbal formula called Jiawei Buzhong Yiqi Decoction curtailed the exorbitant expression of kisspeptin and its receptor.3 That formula was turning kisspeptin activity down, which is the opposite of adding more. In ovariectomised mice, progesterone given alone induced a rise in kisspeptin immunoreactivity, a stain that shows how much of the hormone is present.4 Those are the body's own hormones adjusting the kisspeptin system, rather than a second compound taken alongside a dose. So the record shows kisspeptin sitting in a web of signals that move it up and down. What it does not show is any combination given on purpose and measured as a combination, and a stack built on this literature would be untested in every sense.

Which is better, kisspeptin or PT-141?

No study among the research behind this page compared them, so there is no result to rank. What the kisspeptin side can offer is its own record on sexual function, which is narrower than the question suggests. A 2025 review lists sexual behavior and attraction among the functions kisspeptin is implicated in outside its hypothalamic reproductive roles.5 A 2022 review proposes that chronic use of kisspeptin might help otherwise healthy men with hyposexual drive disorder.6 Hyposexual drive disorder means persistently low sexual desire. The review's own wording is potentially, and a proposal is not the same as a result in men. The only measured sexual-behaviour outcome among these sources is in rats, covered on the the Kisspeptin overview overview. What has been documented about PT-141 is set out on the PT-141 overview. Better would need both in one trial with the same outcome, and nothing of that kind exists here.

Can you take PT-141 and kisspeptin together?

Nothing among the research behind this page gave the two together, in people or in animals. That is a statement about the record, not a finding that the pairing is harmful or harmless. The closest thing to a relevant design is the rat study that put kisspeptin-10 beside a GnRH analogue and yohimbine, with sexual motivation as the outcome. It ran its groups through an open-field reward-proximity chamber, with a female in estrous phase behind a transparent perforated wall for 10 minutes.1 Each rat received a single substance and none received two simultaneously, so even that study measured every compound in isolation. A combination constitutes its own exposure, and its effects are measured as a combination or not at all. For PT-141 and kisspeptin, the honest answer is the second, and what each does separately does not establish what they do together.

Is kisspeptin better than hCG for men?

That comparison has not been made in men in any study among the research behind this page. What can be laid out is where each sits in the chain. Kisspeptin neurons, which coexpress neurokinin B, regulate the activity of GnRH neurons and so the pulsing release of GnRH from the hypothalamus, a 2025 review explains.5 GnRH then drives the pituitary gland, which in turn drives the testes, whereas hCG works at the far end of that chain, on the testes themselves, as covered on the HCG comparison page. Kisspeptin therefore depends on the entire chain responding, while hCG bypasses most of it. The one male animal comparison here is not with hCG at all. In the rat study, intranasal buserelin acetate increased concentration of testosterone but did not affect sexual motivation.1 That rat result is not a ranking against hCG, and it does not carry over to adult men either.

Is kisspeptin a safer IVF trigger than hCG?

That is the claim made for it, and among these sources it rests on review statements rather than on a trial result the reader can inspect. A trigger is the timed injection that ripens eggs before collection. A 2022 review states that kisspeptin can induce a more physiological degree of oocyte maturation that can reduce the risk of potentially life-threatening complications such as ovarian hyperstimulation syndrome seen with human chorionic gonadotrophin.6 Ovarian hyperstimulation is an over-response of the ovaries, and it is the main documented hazard of the hCG trigger, as the HCG safety page sets out. The same review describes kisspeptin in health as inducing normal puberty and modulating ovulation in healthy women.6 Read the scope carefully. Safer here is a comparison about one complication, in women having egg collection. It does not extend to men, to testosterone, or to anyone outside an IVF cycle.

How does native kisspeptin compare with the agonist MVT-602?

This is the only human head-to-head among the research behind this page, and it compared kisspeptin with a longer-acting version of itself. An agonist is a molecule built to switch on the same receptor. After MVT-602, the luteinising hormone rise was similar in women with PCOS and in healthy women, but came earlier in women with hypothalamic amenorrhea.7 Hypothalamic amenorrhea means periods stopping because the brain's signal has gone quiet. In laboratory tests, MVT-602 induced more potent signaling through the kisspeptin receptor and a longer duration of GnRH neuron firing than kisspeptin-54, 115 against 55 minutes.7 So the agonist held the switch on for longer, in cells and in women. That is the point of building it. The study ran in small groups over hours, and it was not built to compare the two on any outcome a person would notice.

Is kisspeptin-10 the same as kisspeptin-54 or kisspeptin-13?

They come from the same gene and act on the same receptor, and they are not interchangeable in the research. Kisspeptin is the peptide product of the KiSS-1 gene and the body's own agonist for the Kiss1 receptor, a 2022 rat study notes.1 The number after the name is the length of the chain in amino acids, the building blocks of a peptide. The human study gave kisspeptin-54, the rat studies gave kisspeptin-10, and the mouse pain study gave kisspeptin-13. That mouse study tested kisspeptin-13 under both physiological and inflammatory conditions, and reported a hyperalgesic effect, meaning heightened pain sensitivity.8 No study among the research behind this page tested two of these lengths against each other. So a result for one length belongs to that length rather than to the family, and a pain finding for kisspeptin-13 cannot be assumed for kisspeptin-10, in either direction. The form people search for is kisspeptin-10, which was never given to a person here.

Is kisspeptin the same as fezolinetant or neurokinin B?

No. They share a circuit, and the approved drug works by blocking a neighbour rather than by adding kisspeptin. A 2025 review says the part neurokinin B plays in menopausal hot flashes led to drugs that block that pathway.5 Antagonism means blocking. In the rat, the cells that carry both kisspeptin and neurokinin B reach into the brain areas that set body heat, and those areas carry the main receptor for neurokinin B.9 A 2025 review of ageing in women lists these blockers among new options set beside standard hormone therapy.10 Fezolinetant is one of those blockers, and its approval is covered on the the Kisspeptin overview overview. A blocker of the next-door receptor and a dose of kisspeptin push the same circuit in different directions, so the approval of one tells you nothing about the other.

In PCOS, how does kisspeptin compare with the signals beside it?

In one human study it was the least connected of the three. Polycystic ovary syndrome, or PCOS, is a common hormone disorder in women, and kisspeptin works alongside two partner signals, neurokinin B and dynorphin. In 110 lean PCOS patients, dynorphin was positively associated with the LH/FSH ratio in both bivariate and pathway analyses.11 The same study found NKB levels were not associated with kisspeptin.11 So the measured hormone pattern tracked a partner signal more closely than kisspeptin itself. In mice the picture runs the other way. The authors of a 2024 mouse PCOS study say their findings complement recent clinical investigations using NKB receptor antagonists to lower LH in PCOS women.12 Those two disagree because one measured levels in women and the other switched neurons off in mice. They are different questions, and neither suggests adding kisspeptin in PCOS.

Do progesterone or oestrogen change how kisspeptin works?

Yes, and that matters for anyone weighing kisspeptin beside other hormones. A 2024 review says progesterone helps control the release and pulsing of the pituitary hormones via its receptors on the kisspeptin, neurokinin B, and dynorphin neurons in the hypothalamus.13 In ovariectomised mice, one group of kisspeptin cells was positively influenced by estradiol, whereas in the arcuate group the administration of estradiol did not affect the system.4 Estradiol is the main form of oestrogen, and the arcuate nucleus is the part of the hypothalamus that holds the pulse generator. So the same hormone moved one cluster of kisspeptin cells and left another alone. In practice, that means kisspeptin does not act in a fixed way. Its effect depends on the hormonal setting it lands in rather than on the amount alone, which no study among the research behind this page tested in a person taking other hormones.

What would a fair comparison of kisspeptin need?

It would need the same kind of people, randomised, measured over the same period on the outcome actually being argued about. The record behind this page contains one human head-to-head, in 21 women over several hours, comparing two versions of the same signal. For the comparisons people search for, nothing of that kind exists here. A fair test against PT-141 would give each to comparable adults and measure the same sexual-function outcome. A fair test against hCG in men would follow testosterone and sperm over weeks. The 2024 PCOS mouse authors point to pharmacological dose-dependent modulation of kisspeptin neuron activity as a possible future therapeutic target.12 That line describes a field still deciding which direction to push the system, which is not the stage at which a winner can be named.
