Kisspeptin is a hormone the body already makes, the brain's starter signal for the reproductive system, and most of the published work measures a person's own level rather than giving them any. What people search for is kisspeptin-10, a short piece of that natural hormone.

A 2021 review describes puberty opening with the initial “kiss” between kisspeptin and the hypothalamic neurons that secrete GnRH. GnRH is the brain hormone that drives the testes and the ovaries. The physiology is well mapped. The outcome data is not. A 2022 review reports that in male reproduction, kisspeptin also plays an important role, getting involved in the regulation of Leydig cells, spermatogenesis, sperm functions and reproductive behaviors. The sharpest administration result is not an encouraging one. In prepubertal male rats, chronic intermittent administration of kisspeptin-10 had a dose-dependent degenerative effect on the plasma testosterone levels and Leydig cells ultrastructure.

Human work exists, and it lives in fertility medicine. A 2020 study gave a receptor agonist and native kisspeptin-54 to healthy women (n = 9), and to women with polycystic ovary syndrome (PCOS; n = 6) or hypothalamic amenorrhea (HA; n = 6). What it tracked was how fast luteinising hormone rose. A 2022 review gives the clinical aim, that kisspeptin can induce a more physiological degree of oocyte maturation during in vitro fertilisation treatment. Those are hormone-timing experiments in small groups, run over hours in a clinic. They are not tests of testosterone, muscle or body weight in anybody.

No kisspeptin product is recorded as approved for any indication among the sources cited on this page. One approval sits close by and is easy to misread. Fezolinetant, a neurokinin 3 receptor antagonist, has recently received Food and Drug Administration (FDA) approval for clinical use. It blocks a neighbouring signal instead of supplying kisspeptin.

What is kisspeptin, and what does it do?

Kisspeptin is a hormone the brain makes, built in the hypothalamus, the part of the brain that sets the pace of the sex hormones. A 2021 review calls kisspeptins a group of polypeptides, recently discovered to play a major role in the regulation of the reproductive axis through influencing GnRH release.1 GnRH is the brain signal that tells the pituitary gland to prod the testes or the ovaries. The chain is short and well mapped, and kisspeptin hits its own receptor first. A 2022 review puts that step plainly: kisspeptin activates the signaling pathway by binding to its receptor kisspeptin receptor (KISS1R) to promote GnRH secretion.2 GnRH then goes out in pulses, and the pituitary answers. The form people search for is kisspeptin-10, a ten-amino-acid piece of the natural hormone, and that is the form most experiments give.

Has kisspeptin been given to people, or only measured in them?

Both, and telling the two apart is the most useful thing on this page. Most of the human work on kisspeptin is a measurement, where someone draws blood and sets the figure beside a diagnosis. Three completed studies on the public trial register have exactly that shape. One is Serum Kisspeptin Levels in Infertile Women.3 Another is Association of Kisspeptin Levels With Insulin Secretion in Diabetes Mellitus.4 A much smaller body of work gives kisspeptin to a person and watches what follows. A fourth entry, Reproductive Hormones During Sustained Administration of Kisspeptin, is listed as RECRUITING.5 A reading tells you where somebody sits, while an administration study tells you what changes when the hormone goes in. Those are different questions, and the evidence behind each one is a different size.

What does a kisspeptin reading actually tell you?

Less than the sheer number of papers measuring it would suggest. One study makes the point better than any summary. PCOS is polycystic ovary syndrome, a common hormone disorder in women of fertile age. A 2024 study measured kisspeptin in 110 lean PCOS patients as subjects, beside the hormone ratio it was meant to explain.6 The result was flat: there was no direct association between kisspeptin and the LH/FSH ratio.6 A 2019 paper on pregnancy shows the other half of the problem. In pregnant mice, blocking the animals' own kisspeptin produced impaired glucose homeostasis.7 GDM is gestational diabetes, the kind that begins in pregnancy. For people, those authors go only as far as a suggestion, that less placental kisspeptin may be a factor in the development of GDM in humans.7 Read that carefully, because blocking a hormone in mice and a careful sentence about people are not the same evidence.

How far has kisspeptin gone in human research?

The clearest human work sits in fertility medicine. A 2020 study in the Journal of Clinical Investigation compared a receptor agonist with the natural hormone. It ran in healthy women and in women with reproductive disorders.8 Those disorders were polycystic ovary syndrome and hypothalamic amenorrhoea, which means periods stopping with no structural cause.8 All three groups were small, and the whole comparison sat inside one study. The reported outcome was timing: luteinising hormone peaked 4.7 hours after the natural peptide and 21.4 hours after the agonist.8 KP54, the comparison arm, is native kisspeptin-54. A 2022 review gives the clinical aim, reporting that kisspeptin can induce a more physiological degree of oocyte maturation during in vitro fertilisation treatment.9 So the human record is real, and it is about hormone timing and egg maturation in a clinic. It was not built to answer what somebody outside that setting wants to know.

Does kisspeptin raise testosterone?

It acts on the switch that controls testosterone, which is not the same as raising it. A 2021 review describes puberty opening with the initial “kiss” between kisspeptin and the hypothalamic neurons that secrete GnRH.10 From there the review is explicit: this pulsatile production of GnRH is followed by a rise in LH and, consequently, in gonadal steroids.10 Gonadal steroids include testosterone, so the pathway is the right one. A 2022 review adds the male side. In male reproduction, kisspeptin also plays an important role, getting involved in the regulation of Leydig cells, spermatogenesis, sperm functions and reproductive behaviors.2 Leydig cells are the testicular cells that make testosterone. What is missing is the measurement a reader wants. A trial in men given kisspeptin, with testosterone followed over weeks, does not appear among the research behind this page.

Does kisspeptin shut testosterone down?

One animal result points the other way. Anybody reading the testosterone claim should read this one beside it. A 2022 study checked the ultrastructure of Leydig cells in prepubertal male rats in response to the administration of a range of kisspeptin doses.11 The injections were repeated rather than given once. Its conclusion was blunt. Chronic intermittent administration of kisspeptin-10 has a dose-dependent degenerative effect on the plasma testosterone levels and Leydig cells ultrastructure in prepubertal male rats.11 That is a rodent experiment in immature animals, and it does not carry over to an adult man. It does show that the direction depends on how the hormone arrives. A single clinic dose and a repeated course are different questions, and only one of them has a human answer.

How does kisspeptin make you feel?

No study among the research behind this page scored mood or libido in people given kisspeptin. The nearest evidence is a small behaviour study in rats. A 2022 report used 50 copulation naive male Wistar rats divided into 5 groups.12 Some got kisspeptin-10 by injection into the belly, and others got it up the nose. The belly route moved both testosterone and sexual motivation, the authors reported.12 The nasal route split the two, lifting sexual motivation while leaving testosterone alone.12 Their own reading of that is the interesting part. Some behavioral effects of kisspeptin can possibly be realized independently of testosterone concentration changes.12 A rat crossing a cage toward a female is a long way from how a person describes feeling, and that distance is the finding here, rather than the rat score itself.

How long does kisspeptin-10 take to work?

Fast, and briefly, which is the practical shape of the molecule. In the 2020 human comparison, luteinising hormone peaked 4.7 hours after the natural peptide and 21.4 hours after the longer-acting agonist.8 Those hours describe a hormone pulse measured in a clinic, and they are not a benefit anybody reported noticing. A 2022 review is direct about the limit, naming a short duration of action and development of tachyphylaxis for the natural forms of kisspeptin.9 Tachyphylaxis means the response fading as dosing carries on, which is why the field has been building analogues instead of using the hormone itself. A peak at hour five also says nothing about what happens over a week, a month or a whole cycle.

Can kisspeptin be taken every day?

No study among the research behind this page reports a schedule for continuous use in people. The reason is worth more than the gap. A 2022 review describes a short duration of action and development of tachyphylaxis.9 Repeating the natural hormone is what the pharmacology argues against, which is different from a question nobody got round to asking. One registered study was built to ask it directly. Reproductive Hormones During Sustained Administration of Kisspeptin is listed as RECRUITING and has not reported.5 The only repeated-administration result with an outcome attached is the rat Leydig-cell work. There, repeated injections produced degeneration in the rats rather than a steady gain.11 Daily use and a single clinic infusion are different questions, and only the second one has a human answer.

Can kisspeptin cause weight gain?

No study among the research behind this page has weighed people given kisspeptin. So the question has to be answered from where the hormone sits, rather than from an outcome. It sits unusually close to appetite and energy signalling. A 2020 review in Nature Reviews Endocrinology says metabolic factors impose a very tight control over kisspeptin synthesis and release.13 The same review goes further, describing the novel role of kisspeptin neurons as active players within the neuronal circuits that govern energy balance.13 In childhood the traffic runs the other way. Leptin, kisspeptin and insulin have been tied to the link between childhood obesity and earlier puberty, a 2022 review reports.14 Both of those describe body weight acting on kisspeptin. Neither describes kisspeptin acting on body weight, and most people asking this have the direction backwards.

Does kisspeptin grow muscle?

Nothing among the sources cited on this page measures muscle, strength or body composition after kisspeptin. That holds for every species in them. This literature was built for another purpose, and the map of it is clear enough to say so. The reviews cited on this page cover puberty, the menstrual cycle, fertility and menopause. A 2025 review in Physiological Reviews lists the span. Kisspeptin and neurokinin B (NKB) play a key role in several physiological processes including in puberty, adult reproductive function including the menstrual cycle, as well as mediating the symptoms of menopause.15 The muscle argument runs through testosterone instead. It inherits every limit of the testosterone evidence above. A hormone that moves luteinising hormone over an afternoon is not the same thing as one shown to build tissue over months.

Does kisspeptin cause hair loss?

Hair is not an outcome in any study among the research behind this page. No review in these sources lists skin or hair among the tissues kisspeptin acts on. The worry is a reasonable one to hold. It follows from the testosterone logic rather than from anything measured. What the reviews do describe is a system sitting upstream of the gonads. Mutations in the KISS1 gene or disorders of the kisspeptin/KISS1R system may lead to clinical symptoms such as idiopathic hypogonadotropic hypogonadism (iHH), central precocious puberty (CPP) and female infertility.2 Those are reproductive outcomes, reported in people born with a genetic fault. None of them is a hair outcome. An untested worry and a documented harm are not the same thing, and this one is still the first.

What is documented about harm?

The documented harms sit in animals, and they are specific enough to name. Repeated kisspeptin-10 in prepubertal male rats had a dose-dependent degenerative effect on the plasma testosterone levels and Leydig cells ultrastructure.11 That is a structural change, not a passing dip. A 2025 mouse study points somewhere unexpected. Different doses of KP-13 were administered to adult male C57BL/6 mice.16 In the formalin test, the KP-13-treated mice showed increased nocifensive behavior compared to the control group.16 Nocifensive behaviour is what an animal does to guard itself from pain. The authors concluded that KP-13 has a hyperalgesic effect, meaning the mice felt pain more readily rather than less.16 Human harm data is thinner, because the clinical work was built around hormone responses in small groups rather than around tolerability over time. An absence of reported problems across a handful of single-dose studies is not a safety record.

Is kisspeptin approved for anything?

No kisspeptin product is recorded as approved for any indication among the sources cited on this page. One nearby approval exists, and it is easy to misread. A 2025 review notes that a neurokinin 3 receptor blocker has recently received Food and Drug Administration (FDA) approval for clinical use.15 That drug is called fezolinetant.15 It blocks a nearby signal, and it does not add kisspeptin to anyone. Work on this pathway is live all the same. A 2023 review in BMJ Medicine describes new work aimed at the kisspeptin axis, just above the GnRH signal.17 An approved drug acting on the same circuit is not an approval for kisspeptin itself. That gap matters when somebody cites one to justify the other, because they are different claims about different molecules.

How is kisspeptin thought to work?

The mechanism is better established than almost anything about the outcomes. It is a pacing system rather than a fuel. A 2016 review in Nature Reviews Endocrinology places the hardware precisely. The arcuate nucleus kisspeptin neurons that innervate the projections of GnRH neurons in and around their neurosecretory zone are key components of the pulse generator in all mammals.18 The pulse generator is the clock that decides how often GnRH is released. The rhythm matters as much as the amount. Steering that clock downward works too. In a 2024 experiment, targeted inhibition of endogenous kisspeptin neuron activity in a mouse model of PCOS reduced the abnormally hyperactive LH pulse secretion and hyperandrogenemia to healthy control levels.19 That is the cleanest sign that the circuit is doing the driving. Its authors add the caveat themselves. Mouse models can offer mechanistic insights into neuroendocrine processes in PCOS-like conditions but may not perfectly mirror PCOS in women.19

What we don’t know

The gaps in the evidence matter as much as the findings.

  1. 01What kisspeptin does to testosterone in adult men over time. The human administration work among the research behind this page measured luteinising hormone in women over hours, and the only repeated-administration result is a rat study in immature animals.
  2. 02Whether a measured level can be acted on. Studies in these sources report kisspeptin readings beside a diagnosis, and none of them tests whether moving the figure changes anything.
  3. 03What continuous use would look like. The one registered study of sustained administration is listed as recruiting and has not reported.
  4. 04What material obtained outside a clinic contains. No source among the research behind this page characterises the purity or composition of kisspeptin sold under that name.
  5. 05Whether the rodent Leydig-cell degeneration has any counterpart in people. It was reported in prepubertal rats given repeated injections, and nothing in these sources follows it up in an adult of any species.
  6. 06How kisspeptin behaves alongside other compounds. The combinations documented among the sources cited on this page are laboratory pairings inside animal disease models.
  7. 07Anything about hair, muscle or body composition. Those outcomes are not measured in any study among the research behind this page.
