Melanotan protocols in the published literature

No source in this record sets out a dosing schedule for Melanotan-II, and the figures traded under that name have no study behind them. The literature holds a handful of experimental protocols, nearly all from the 1990s, run on about thirty people. Melanotan-I and Melanotan-II are separate molecules, so a figure attached to one says nothing about the other.

The fullest human protocol is a 1996 phase I pilot. It was a single-blind, alternating day (saline or MT-II), placebo-controlled trial conducted in 3 normal male volunteers at the starting dose of 0.01 mg/kg of MT-II, injected under the skin from Monday to Friday for two weeks. Two subjects were escalated by 0.005 mg/kg increments to 0.03 mg/kg and one to 0.025 mg/kg. Three men, thirty years ago, is the foundation under every figure circulating today.

The other two human studies measured something else. Seven volunteers received Melanotan-I at 0.16 mg/kg/day across two weeks, read for pigment on day 14 and day 21. Twenty men with erectile dysfunction took Melanotan-II in a crossover design, where at a dose of 0.025 mg/kg, 12.9% of subjects had severe nausea. None of the three followed anyone past three weeks.

Neither molecule is a licensed medicine, so no vial carries a labelled amount a reader could work from. FDA records hold three Class II recalls of compounded melanotan injections — a 10 mg Melanotan II presentation, a 1 mg/mL ten-millilitre vial, and Melanotan I at 200 mcg/mL — every one for a sterility failure. A 2018 analytical review lists melanotan II among the image-enhancing polypeptides that have been falsified.

What is the proper dosing for Melanotan 2?

There is not one. No study in this record has set out to establish the amount of Melanotan-II a person should use for tanning, and the three human investigations on record were each designed to answer something narrower. What exists in place of a protocol is a dose escalation from 1996. A single-blind, alternating day (saline or MT-II), placebo-controlled trial was conducted in 3 normal male volunteers at the starting dose of 0.01 mg/kg of MT-II.11 The team then worked upward, and two subjects were escalated by 0.005 mg/kg increments to 0.03 mg/kg and one to 0.025 mg/kg.11 Read that as a tolerability exercise rather than a schedule anybody arrived at, because an escalation design exists to identify the step at which unwanted effects appear, which is a separate question from what amount produces pigment. The authors closed with a figure of their own, and its wording carries the entire distinction: the recommended single MT-II dose for future Phase I studies is 0.025 mg/kg/day.11 That proposes an amount for the following experiment, not for a person at home.

How much Melanotan 2 was injected in the published studies?

Every figure in the human record is expressed per kilogram of body weight, which is how an investigation records an amount, not how a vial is sold. The 1996 pilot opened at the starting dose of 0.01 mg/kg of MT-II and its highest step was 0.03 mg/kg.11 The 2000 crossover study in men with erectile dysfunction settled on a different figure, and reported that at a dose of 0.025 mg/kg, 12.9% of subjects had severe nausea.1 This page publishes no conversion from those figures to a quantity in milligrams, and the reason is not caution for its own sake. A per-kilogram figure carries its investigation's conditions along with it: a known substance of established concentration, volunteers screened before entry, and somebody observing afterwards. Multiplying it by a reader's body weight removes all of those and leaves an arithmetic result wearing a study's authority.

How much Melanotan 2 a week, and how often per day?

Frequency can be read straight off the page, because the 1996 team wrote the schedule out in full. Subcutaneous injections of MT-II or saline were given daily (Monday-Friday) for 2 consecutive weeks.11 That is five shot days a week with the weekend left out, and the design swapped real material and saline on back-to-back days, so each man was given MT-II on about every second weekday. The authors summed the course up as tanning activity in humans given only 5 low doses every other day by subcutaneous injection.11 Five doses across two weeks is a long way from the daily routine the question assumes. No study covered here sets a daily schedule against an every-other-day one in the same men, so the gap between doses is a trait of one small study rather than a finding about what any gap achieves.

What was the Melanotan 1 protocol for tanning?

Melanotan-I has the one human protocol that was built around skin colour from the start. Its figures sit well above anything on record for Melanotan-II. Seven normal volunteers (six males and one female) with tanning skin types III or IV (Fitzpatrick scale) were given 10 daily subcutaneous injections in a 2000 study.2 The Fitzpatrick scale ranks how fast skin burns or darkens. The amount was fixed rather than stepped up, and the agent was administered at a dose of 0.16 mg/kg/day (Monday-Friday), over a two week period.2 The gap between 0.16 mg/kg for Melanotan-I and 0.03 mg/kg for Melanotan-II is not a difference in strength that anyone has measured. They are two molecules, given to two sets of volunteers, for two endpoints, by teams who never set them side by side.

How long does it take for Melanotan 2 to make you tan?

The earliest tanning measurement anybody published came three weeks after the first injection, and it is a measurement window rather than an onset. No study in this record of either molecule recorded when colour first showed, and the reason sits in the dosing schedules. In the 1996 pilot, two of the three volunteers had increased pigmentation in the face, upper body and buttock, as measured by quantitative reflectance and by visual perception 1 week after MT-II dosing ended, which puts that reading three weeks after the course began.11 The Melanotan-I study ran the same way, comparing baseline to the end of the daily dosing period, (day 14), and then to day 21.2 So three weeks is the first date on which anyone looked, not the date anything started. The question people are typing, which is how many days until this shows, has not been asked in a published study of either molecule, because nobody read a volunteer on day three, day six or day nine. A figure in days quoted anywhere else is an impression rather than a measurement of skin.

Is there a Melanotan dosage chart?

Not a published one. No regulator, trial or maker has issued a table of amounts for either molecule, and nothing in this record could be set out as one without inventing the rows. What could fairly be tabulated is thin enough to state in a sentence: three human studies, two molecules, four distinct amounts, one route and two schedules, with each of those studies enrolling fewer than twenty-one people. A chart implies a map from some input, usually body weight or a goal, to an output amount. Nothing in this record supports that map, because the only escalation on record ran upward until the 0.03 mg/kg dose produced Grade II somnolence and fatigue in one of two subjects (WHO standards), and stopped at that step.11 A table built on a stopping point decorates a figure that was never a target rather than summarising anything measured.

What does a 10 mg Melanotan 2 vial contain?

The 10 mg figure people search for describes a container rather than an amount anybody was given at once, and it states how much dry material was sealed into the vial before any liquid was introduced. FDA enforcement records document three presentations under this name. One recall covers Melanotan II, 10mg injection from a Florida compounding pharmacy.12 A second covers Melanotan II 1 mg/mL (10 mL) Injection, 10mL vials, which is ten milligrams of material distributed through ten millilitres of liquid.13 A third covers Melanotan I 200mcg/mL (2mg/ml), a) 1.5mL-vial, b) 5 mL-vial.14 Those are the only concentrations here from a documented product rather than an investigation, and all three were withdrawn for a sterility failure, which describes how a batch was made rather than the molecule inside it. A vial size tells a reader what they are holding and nothing about what to do with it.

How do you reconstitute a melanotan vial?

Reconstitution is arithmetic performed on two numbers the vial and the syringe already supply, and it is the one part of this subject with a definite answer. The two inputs are the stated quantity of dry material and the volume of bacteriostatic water introduced into the vial. Dividing the first by the second produces the concentration of the liquid. A vial stated at 10 mg, reconstituted with 2 mL of water, holds 5 mg in every millilitre, identical to 5,000 mcg per millilitre, because one milligram is a thousand micrograms. Alter the water and the concentration alters with it, while the quantity of material stays where it was. That calculation tells a reader what is present in a given volume of liquid. It does not tell them what volume to draw, and nothing in this record about either molecule establishes that figure. The recall records carry a Melanotan II 1 mg/mL (10 mL) Injection and a Melanotan I 200mcg/mL (2mg/ml) presentation, so an identical marking on a syringe can represent different quantities from one vial to the next.1314

How do mg, mcg and syringe units relate?

Three units perform different jobs, and confusing them is where the arithmetic ordinarily goes wrong. Milligrams and micrograms measure the quantity of peptide, millilitres measure the liquid carrying it, and the markings on an insulin syringe measure neither one directly. One milligram is a thousand micrograms, so a concentration written as 2 mg/mL is identical to one written as 2,000 mcg/mL. A U-100 insulin syringe is divided into 100 units across a single millilitre, which makes each unit one hundredth of a millilitre. Converting units into a quantity therefore means multiplying that fraction of a millilitre by the concentration of the reconstituted vial. The step people omit is that a syringe unit measures a volume and never a quantity. The recall records hold both a Melanotan II 1 mg/mL (10 mL) Injection and a Melanotan I 200mcg/mL (2mg/ml) presentation, so ten units out of one vial is not ten units out of the other.1314

How were the melanotan injections given in the studies?

Every human protocol on record used a shot under the skin. None of them used any other route. In the 1996 pilot, subcutaneous injections of MT-II or saline were given daily, and the 2000 Melanotan-I study gave 10 daily subcutaneous injections to seven volunteers.112 The animal work used other routes, and that is worth keeping apart rather than folding in. A 2006 rat study delivered melanotan-II i.v. (0.1, 0.3 and 1 mg/kg) and also straight into a small control centre deep in the brain.3 A 2018 mouse study used intraperitoneal administration, meaning a shot into the belly, and found that it causes a profound, transient hypometabolism and hypothermia.7 Those last two words mean a drop in body heat and in the rate the body burns fuel. Routes do not substitute for one another: material placed in the brain, the belly or a vein reaches tissue at concentrations a shot under the skin does not match, and nothing in this record converts between them for either molecule.

What happened to people at the amounts the studies used?

This is the fullest part of the dosing record, and it runs the opposite way from the question people bring to it. At the top of the 1996 escalation, the 0.03 mg/kg dose produced Grade II somnolence and fatigue in one of two subjects (WHO standards).11 Lower down the range, mild nausea, not requiring antiemetic treatment, was reported at most MT-II dose levels.11 In the 2000 crossover study, at a dose of 0.025 mg/kg, 12.9% of subjects had severe nausea, which is about one dose in eight, in volunteers screened beforehand and watched throughout.1 Erections tracked the amount rather than cropping up at random. The 1996 team described spontaneous, penile erections which were intermittently experienced for 1-5 hours after MT-II dosing, depending on the MT-II dose.11 So the published figures are not amounts at which nothing happened; they are the amounts at which the recorded effects were judged acceptable inside a supervised study, which is a different standard from the one a reader applies alone.

Why do study amounts not transfer to a vial bought online?

Because the published figures describe a known substance, and an unlabelled vial is not one. The link between them is an assumption, and it has been tested often enough to know how it fails. Neither molecule is a licensed medicine. A 2012 report describes injectable synthetic melanotropic peptides (often called melanotan) to enhance tanning as available over the Internet despite being unlicensed compounds with an unproven safety record.5 A 2014 report adds that Melanotan-II is unlicensed and incompletely tested, the extent and types of adverse effects are unknown.6 FDA records hold three Class II recalls of compounded melanotan injections, including one covering Melanotan II, 10mg injection, every one for a sterility failure.12 The vials have been checked too. A 2018 review of falsified peptides lists melanotan II among them, and records that multiple reports have namely described the presence of the wrong active pharmaceutical ingredient (API), the wrong dosage or the absence of the API.8 Put those together and the arithmetic falls apart. A figure in milligrams per kilogram assumes the vial holds the stated substance, in the stated amount, with nothing else in it, and not one of those three has been shown for material sold under this name.

How much does Melanotan 2 cost?

No source in this record records a price, and that absence tells a reader something rather than merely getting in the way. What the literature does document is where the material turns up. A 2022 Dutch case report notes that Melanotan II, also known as 'Barbie drugs', can be purchased over the internet and in gyms to facilitate sunless tanning.9 A 2025 history of tanning technologies adds that tanning injections introduced a new host of health risks for twenty-first-century consumers.10 Price is easy to find and tells a reader nothing, because a seller sets it rather than a study measuring it. The numbers that would genuinely inform a purchase are what the vial holds, at what concentration, and under what manufacturing conditions, and those are the figures no analysis in this record of this supply has confirmed.

Why do the reported amounts vary so much?

Four reasons, and none of them is that a particular figure is more correct than the others. The first is the molecule, since Melanotan-I was administered at a dose of 0.16 mg/kg/day while Melanotan-II was given at a fraction of that, and no study we cite ran both in the same people.2 The second is what was measured. The 1996 escalation hunted for the step at which side effects appeared, the Melanotan-I study read skin colour with a light meter, and in the 2000 crossover study penile rigidity was monitored for 6 h using RigiScan.1 Three questions produced three amounts. The third is species. Rodent work used figures reaching 1 mg/kg by routes no person uses, and a 2007 mouse study confirmed the low penetrability of the peptide in mouse brain at those amounts, with nothing in these sources turning an animal figure into a human one for either molecule.4 The fourth is the calendar. All the human work finished before 2001, and a Phase 2 trial of Melanotan II (MT-II) as an Adjunct to NB-UVB Phototherapy for Repigmentation in Stable Nonsegmental Vitiligo is recruiting rather than reporting, with the published additions across the quarter-century between them being case reports.15

What we don’t know

The gaps in the evidence matter as much as the findings.

  1. 01What amount produces a tan. The only escalation on record was designed to find where unwanted effects began, which is a different question, and it stopped as soon as it found them.
  2. 02How many days pass before pigment appears. Both tanning investigations read their results on dates fixed before the course started, so the earliest reading either one can report is the date it happened to look.
  3. 03Whether a daily schedule, an alternating one, a cycle or a break changes anything. No study in this record has compared two intervals of either molecule, so there is no schedule to set against another.
  4. 04Whether anything above 0.03 mg/kg of Melanotan-II has been given to a person deliberately. The published escalation stopped at that step and no study among the research behind this page has published a higher one.
  5. 05What either molecule does past three weeks. The longest documented human course is two weeks of injections with a follow-up reading seven days later.
  6. 06How the two molecules relate on amount. Melanotan-I was given at roughly five times the Melanotan-II figures, and no investigator has run them side by side.
  7. 07What is inside a vial sold under this name. Falsified peptide preparations have been documented to contain the wrong ingredient, the wrong quantity, or none of the stated ingredient.
