Matrixyl: what the research measured

Matrixyl (palmitoyl pentapeptide-4) has one measured benefit in people that stands on its own: fewer visible fine lines and wrinkles after 12 weeks, compared with the same cream without it, in 93 women. Its other reported benefits, more collagen and faster wound healing, were measured in cells and animals.

The human result was scored two ways. The peptide side showed a significant improvement over the placebo side by computer image analysis and by expert graders, which is stronger than either method alone. A smaller 2023 trial in 21 Indonesian women pointed the same way over eight weeks.

The laboratory and animal benefits are wider and less certain to transfer. In animals with skin wounds, Matrixyl patches and creams improved wound healing by 63.5% to 81.81% against untreated controls, and in photo-aged mouse skin a carrier system holding the peptide reduced wrinkles. Those are different tissues, under different conditions, from an ageing face.

What has not been measured matters as much. Among the studies behind this page, nobody counted collagen in human facial skin after using Matrixyl, nobody followed results past 12 weeks, and every trial summary describes women only.

What does Matrixyl do to your face?

In the one trial built to answer this cleanly, it reduced wrinkles and fine lines more than an identical cream without it. That 2005 trial used a split-face design, 93 women and 12 weeks of use, and it judged the result in two separate ways. Computer analysis scored the photographs, trained graders rated the same images by eye, and both found the peptide side ahead.

The women's own view agreed, more loosely. In self-assessment, they reported significant improvement in fine lines and wrinkles, and only directional effects, meaning a trend short of significance, for other aspects of facial appearance.1 So the measured benefit is specific: lines and wrinkles, rather than pigmentation, texture or firmness.

What the trial did not do is equally specific. Its summary did not report the magnitude of the change, the investigators did not collect skin biopsies, and nobody was followed after the 12 weeks concluded. The result establishes that this peptide, at this concentration, in this particular moisturiser, outperformed the moisturiser alone in women. It does not tell a reader whether the difference is large enough to notice in a mirror.

How long does it take to see results from Matrixyl?

The human trials examined several time points, and none of them investigated how early a single-peptide cream begins working. The 93-woman trial reported its result at 12 weeks, and the 2023 trial at eight weeks. Only the combination products were evaluated earlier.

The 2009 eye-area studies in female volunteers measured periorbital skin roughness with a three-dimensional optical scanner at two and four weeks3, and three of the products significantly improved crow's feet smoothness after four weeks, compared with untreated skin3. Those products also contained niacinamide and other actives, so the four-week result belongs to the combination.

A 2026 case report recorded measurements at six and twelve weeks in one woman applying a Matrixyl serum, and documented changes on both sides of her face by the end. The accurate summary is that changes were measurable at four weeks for combinations and at eight to twelve weeks for the peptide individually, and that the earlier weeks were never examined for Pal-KTTKS alone among these sources.

Does Matrixyl work on crow's feet?

It is the best-tested area of the face, and the evidence is positive but small. The 2023 trial measured its three groups with instruments for skin moisture, water loss and elasticity, along with photographs and a crow's feet grading scale4, and it found improvement in several participants using either peptide4, with the Matrixyl peptide ahead of the placebo.

The 2009 blends add a pooled result. When the two studies were combined, all three products tested in both were significantly better than no treatment at smoothing periorbital skin after four weeks.3 The comparison there was untreated skin, not a placebo cream, which is a weaker control because any moisturiser can make skin look smoother.

Read together, both point the same way, and neither is large. The 2023 trial split 21 women across three groups and called itself an initial study, and the 2009 products cannot separate the peptide from the niacinamide beside it.

Does Matrixyl increase collagen?

In cultured cells, mostly yes; in human skin, it has not been measured among these sources. The cell results are the foundation of every collagen claim on a label, and they are not perfectly consistent.

A 2007 study in rat tendon cells found KTTKS raised type I collagen and slowed the breakdown of the genetic message that codes for it, alongside a rise in TGF-beta, a growth signal that drives collagen production.5 Most of the laboratory work behind this page reports an increase of some kind.

Not all of it does. A 2019 in vitro study of Matrixyl-like peptides found that three of them showed no relationship between concentration and collagen production6, although a genuine effect would ordinarily produce one. In people, the 2024 Matrixyl 3000 eye-cream trial measured collagen density by ultrasound, but the summary we could read does not report that result. So the accurate reading is a largely consistent cell signal with exceptions, and a cell signal is not the same as collagen measured in a human face.

How does Matrixyl switch on collagen production?

The cell studies point to two routes, both still at the theory stage. The first is the growth-signal route described above: the peptide raises TGF-beta, which tells fibroblasts to make more collagen and to keep the instructions for it intact for longer.

The second involves cell energy. A 2013 lab study found that a mix of niacinamide, Pal-KTTKS and an olive-oil compound raised the energy level of adult skin fibroblasts in parallel with markers of skin structure7, bringing them closer to the levels observed in cells from newborns. That was a combination, and niacinamide is itself a precursor of the energy molecule being measured, so the peptide's individual contribution cannot be separated.

Neither mechanism has been traced in living human skin among the studies behind this page. They describe plausible biological machinery, which is valuable for understanding the claim, and they do not establish that the machinery operates in a face beneath a cream.

What does Matrixyl 3000 do for skin?

The 3000 pair, palmitoyl tripeptide-1 and palmitoyl tetrapeptide-7, has been tested mostly inside larger formulas, so its benefits are harder to isolate than the original's. The 2024 eye-cream trial assessed moisture, elasticity, facial imaging and collagen density with four different instruments8 over 12 weeks, and its authors reported broad improvement in the eye area. The cream also held a yeast and rice ferment and a sugar compound.

In skin kept alive outside the body, a 2026 study found that a peptide mix including both 3000 peptides raised levels of type I, III and IV collagen9. That mix also held two other peptides.

A 2015 methods study tested a blend of palmitoyl oligopeptide and palmitoyl tetrapeptide-7 against a placebo. By ultrasound, the blend reduced the thickness of a low-density band beneath the epidermis10 in this in vivo work, one of the ageing markers the study set out to track. Its summary does not say on whom the test was run, which limits what can be made of it.

Does Matrixyl need help getting into the skin?

Probably, and a large share of the research is about giving it that help. A 2022 review concluded that the permeability of anti-wrinkle peptides can be raised to useful levels with various enhancement methods11, and it counted KTTKS among the peptides where this had worked.

The methods tested on Matrixyl are physical and chemical. On donated human skin, microneedles raised the signal from Pal-KTTKS and two other peptides by 2 to 22 times12, measured by fluorescence rather than by amount. A 2025 animal study found that micelles made from a liquorice-derived salt significantly increased how much of the peptide crossed into and stayed in the skin13.

These are delivery experiments rather than wrinkle trials. They show the barrier can be crossed more easily, and in doing so they confirm that an ordinary cream leaves most of the peptide on the surface. Whether better delivery produces a better face is a separate question that none of these studies tested in people.

What has Matrixyl been shown to do in animals?

The animal work covers two areas, sun-damaged skin and wounds, and both used the peptide inside a delivery system. In the sun-damage work in animal models, a 2025 study reported that its peptide-loaded micelles boosted collagen and hyaluronic acid in cells and in photo-aged animals13, and it described reduced inflammation and wrinkles alongside better elasticity13.

That is a strong result for the system rather than for the peptide, since the carrier itself was built from bioactive plant compounds. Photo-aging in an animal is produced on purpose with ultraviolet light over weeks, which is a model of sun damage and not a copy of decades of human ageing.

The wound results are covered in the next section. In both areas, the step from a treated animal to a person using a shop-bought cream has not been taken among these sources.

Does Matrixyl help wounds heal?

In animals, yes, across several experimental designs, and none of them involved a human wound. The 2022 study behind the 63.5% to 81.81% figures compared Matrixyl patches and creams with an untreated group, a blank patch, a blank cream and a commercial dressing over 21 days, with animals randomly allocated to seven groups.

In a 2024 rat burn study, dressings enriched with Pal-KTTKS healed significantly better than the control dressing14, with the best results when stem cells were added. A 2024 study found that new versions of the peptide with ring-shaped fatty tails healed wounds in diabetic rats15.

The pattern deserves careful reading. The strongest wound results come from modified molecules, stem-cell dressings, or the peptide incorporated into a patch, rather than from Matrixyl in a cream by itself. They demonstrate that the collagen fragment can improve a dressing; they do not demonstrate that a cosmetic serum assists a cut or a surgical scar.

Do people using Matrixyl notice a difference themselves?

Some do, and the self-reported results are softer than the measured ones. In the 93-woman trial, participants rated their own fine lines as significantly improved, while changes in other aspects of their appearance were only directional. The 2023 trial also administered a self-assessment questionnaire, and its summary reports the Matrixyl peptide ahead of acetyl hexapeptide-3 on that questionnaire as well as on the instruments.

The case report gives a sense of scale for a blended serum. After 12 weeks, the skin-analysis software rated the serum-only side of her face one year younger, against five years on the side that also received a light and current device16.

One year of estimated skin age, in one individual, with a serum that also contained hyaluronic acid and niacinamide, is a modest signal. Self-assessment is useful for knowing whether a change can be seen, and it is also easily swayed by what people expect, which is why the split-face, double-blind design of the 2005 trial carries more weight.

How large is the Matrixyl effect?

The summaries mostly do not say, which is itself the useful fact. The 2005 trial reports a significant improvement over placebo without providing a percentage in its abstract, and the 2023 trial reports superior results without figures. Statistical significance means a difference is unlikely to be chance; it does not mean the difference is large.

The 2023 authors themselves called for further study with a more adequate number of participants and a longer duration4, which is a fair description of where the size question stands.

The large numbers in circulation come from other settings. The 63.5% to 81.81% figures describe wound healing in animals, and percentages from combination creams belong to those combinations. A precise figure for Matrixyl's wrinkle reduction deserves the question of which study produced it, because none of the human trial summaries behind this page provides one for the peptide individually.

Which hoped-for Matrixyl benefits have not been measured?

Several of the benefits most frequently claimed for it lie outside the evidence. Lifting or tightening, smaller pores, scar reduction, durable results after discontinuation, and any effect in men do not appear as measured outcomes in the human trials behind this page. Neither does collagen quantified in a human skin sample after applying Matrixyl.

The delivery problem is the reason the gap matters. The 2022 review warned that most anti-wrinkle peptides might not reach their targets in the skin at the right concentrations11 without help, and the collagen claim depends on exactly that.

None of this means the missing benefits are false. It means they have not been investigated in a trial of the peptide, which is a different situation from having been tested and failed. The fine-line result is the portion that has been examined and answered, and the documented harms are catalogued on the Matrixyl safety page.

What we don’t know

The gaps in the evidence matter as much as the findings.
